On their website it says their printer outputs at 250 ppi. Is the ppi set according to the lab?

Oh, that's excellent! Yes, you should use 250ppi.

YOU do it, not the lab.  Choose your Crop Tool, enter "20in" and "30in" for the size, and "250" for the resolution, then crop.  Then sharpen.

I beg you not to print such a large photo until you've taken the Sharpening Class.
